Daffodil Chromoplast DNA: Comparison with Chloroplast DNA, Physi cal Map, and Gene Localization
نویسنده
چکیده
In daffodil, development of chloroplasts or chromoplasts is not accompanied by plastid DNA modification. This has been shown by comparing the fragment pattern of different restriction endonucleases, and by the lack of methylation of CCGG sequences. A physical map has been constructed for the plastome using the restriction endonucleases Sal I, Pst I, and Bgl I. The fragments containing the genes for the large subunit of ribulose bisphosphate carboxylase/oxygen ase (rbcL), the alpha, beta , and epsilon subunits of the ATP synthase complex (atpA, atpB, atpE), cy tochrom e/(petA ), and for the 51 kDa chlorophyll apoprotein of photosystem II (psbB) have been identified. The respective gene locations correspond to those on spinach chloroplast DNA.

Apparent Identity of Chromoplast and Chloroplast DNA in the Daffodil, Narcissus pseudonarcissus
Although chromoplasts and chloroplasts are mor phologically and functionally very different, it has been shown in certain plants and under certain con ditions, that they are interconvertible [see 1 5]. One could then expect such plastids to contain, at least qualitatively, the same plastome. It appears questionable, however, whether this applies to all plastid types. For example, FreyWyssling...

Background: Gossypium thurberi is a wild diploid species that has been used to improve cultivated allotetraploid cotton. G. thurberi belongs to D genome, which is an important wild bio-source for the cotton breeding and genetic research. To a certain degree, chloroplast DNA sequence information are a versatile tool for species identification and phylogenetic implications in plants. Different ch...

Previous studies have indicated that in all land plants examined to date, the chloroplast gene trnLUAA isinterrupted by a single group I intron ranging from 250 to over 1400 bp. The parasitic Epifagus virginiana haslost, however, the entire gene. We report that the intron is missing from the chloroplast genome of twoarctic species of the legume genus Hedysarum (H. alpinum, H. ...

The chloroplastic DNA of 64 accessions of apple (Malus spp.) (54 Iranian genotypes, five commercial cultivars and five rootstocks) were analyzed to reveal their haplotypes by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method. Approximately 4320 bp of the chloroplast genome was analyzed, by using two chloroplast universal primer pairs and two restriction ...
